Round 1

Reviewer 1 Report

At the end of sentences put the references before de final dot.

Line 66. The sentence “Samples were collected in the Tlalpan Mayoralty.” Is a matter of methodology, should not be in the introduction. The same for the sentence “but the participants in this study came from urban areas” in line 69, “The samples from northern Mexico were collected in the state of Chihuahua” in line 71, “Samples from this population were obtained in Chihuahua City” in line 73.

A map with the location of the population included in the study should be provided.

Line 117, when describing PCR-SSP methodology, please provide a citation.

Line 141, This sentence in result section is a matter of methodology: “Firstly, we made a whole-population frequencies analysis (Mexican-mestizo, MM, n=502).”

Line 154, authors mention that “The estimates of Hardy-Weinberg show a deviation from expected/observed heterozygosity in… HLA-DRB1 (Obs. Het. = 0.9143/Exp. Het. = 0.9329; p-value= 0.104) and HLA-DQB1 (Obs. Het. = 0.8327/Exp. Het. = 0.8186; p-value= 0.676)”, but p-value is higher than 0.05. Also, in other populations form table 1, p-value is not significant for several genes.

Author Response

Thank you very much for your attention and the opportunity to improve our manuscript.

Author response: The lines describing were the populations come from were moved.

A map with the location of the population included in the study should be provided.

Author response: A map was made showing the location of the three populations studied.

Line 117, when describing PCR-SSP methodology, please provide a citation.

Author response: The citations on which our methodology was based were placed.

Line 141, This sentence in result section is a matter of methodology: “Firstly, we made a whole-population frequencies analysis (Mexican-mestizo, MM, n=502).”

Author response: the idea was rewritten to improve understanding of the paragraph

Line 154, authors mention that “The estimates of Hardy-Weinberg show a deviation from expected/observed heterozygosity in… HLA-DRB1 (Obs. Het. = 0.9143/Exp. Author response: the description of these results was corrected based on table 1.

Reviewer 2 Report

The manuscript is well written and clear to understand. I don't have any major criticism to make. However, to improve the clarity of data I have made the following suggestions:

1) In Table 1, there are many p-values. It will highlight the data more by putting * next to all the significant p-values.

2) Although AF and HF are defined under the tables. They should be spelled out in full the first time mentioning them in line 174 and 239.

Thanks a lot for the suggestion, now is corrected.
